ELISAs involve a minimum of one antibody with specificity for a specific analyte or antigen and constitutes the basic protocol listed below:

A liquid sample with the target analyte is added to a microwell that is pre-coated with analyte-specific antibodies. Any target analyte in the sample gets attached to the antibodies.

An enzyme-linked antigen or antibody is added to the microwell, also attaching to the analyte-specific antibodies coated to the microwell.

A substrate is added which modifies its color as it communicates with the enzymes linked to the antibodies or analytes.

The color change is quantified with a spectrophotometer.

Quantitation of the analyte is found by making a comparison of the test sample results with a standard curve of well-known analyte concentrations.
